DotA is a very popular custom map for Warcraft 3. It is based on Aeon of Strife (AoS), a Custom Map for Starcraft 1.
The general concept would be:
2 Teams with one base each
autospawning, computer-controlled creeps fight between those bases, usually separated to multiple lanes.
players pick one hero to support either side in killing the enemy base. This involves killing creeps, which usually provide experience and money and killing the enemy team's heroes to destroy the enemy base unhindered.
There are a huge amount of maps and stand-alone games (Storm of the Imperial Sanctum, Heroes of Newearth, League of Legends), which follow the same concept. Those are generally referred to as AoS, DotA or MOBA (Massive Online Battle Arena) maps/games, while DotA and AoS usually refers to the "classic" game style involving creeps, while MOBA also includes similar games without a base or creeps (Smashcraft, Bloodline Champions).

PVP Arena could be an appropiate description for the 2nd type, so MOBA would be appropiate as well.
RPG maps are definitely not DotA maps.
Tug of War shares some similarities; however in a Tug usually the creep spawning is player-controlled via spawning buildings, and you usually just control a builder unit and there are no heroes directly fighting each other, while in an AoS map, players have little or no influence on the creeps (for DotA you are able to destroy key enemy buildings to upgrade your team's creeps, or use auras and buff spells to improve them with your hero).
DotA stands for Defence of the Ancients. People generally try to piggyback on the success of DotA by saying 'My map is a bit like DotA but without the creep waves and buildings and with RPG elements'. Aka, a team PvP arena with a leveling system and equipable items.
