Not completely, but you can save a boolean value in a bank and set it to true when the map starts. And when the map start check if this boolean is true, if so end the game for player. But ofcourse the bank file can be deleted.
Perhaps it can be done through banks or such? Set up a system where the map will close itself if any banks are found to have been created upon the players entering it, then make it so that the first players who join create a bank. Something in that direction. You'll need someone with more proper data knowledge for details, I guess.
Can I make it so that my map can only be played once? Ever. Like ever.
Not really. I don't know why you'd ever want to do this, but you could create bank files on the player's computer and then check for their existence. If they're found, end the game. But a smart player can find those files and delete them all.

I was thinking more along the lines of a random bank system that would save a random bank among the possible banks to save it too. Then if they have one of those possible banks, end it.
Because you can truly find out who liked the map if they're willing to kill the bank file again and again, just to play. I might actually set it up so that there are 2 separate sections in the bank, one contains the stats of the player and the other one contains the info that they've played before. Also it's the ultimate troll against the popularity system. Are you afraid to play a map that you can only play once. Also it would be hilarious if it became the most popular map.

Why would you want to make a map that no one can ever play again?
