You could store them in an varriable or array if you have multiple players and when a supply depot is being build then kill the unit that is stored in the variable/array.
@Selfcreation: Go You can make it so when the ability to create thetower is used it runs a search and destroys other towers for that player, no triggers needed.
You might want to try it with the behavior type Spawn, there you can set which unit to spawn and a limit, as soon as the limit is reached it will destroy the oldest spawn.

I was wondering if there is a way to track for example:
SCV makes 1 supply deppot , well if it makes a second one it destorys the first one,
how would i track the ammount of specific buildings being build?

well what it is really is this:
I have Heroes that have spells AND towers , but if they buy enought *Energy Potion* they can just Spam Towers. witch makes it to easy!!
so i want to limit them to 1 tower. I have 5 heroes and each have 1 tower...
so 5 towers for up to 5 players.
do I need to make an ARRAY for each tower?

well i got it to work , what i did was.
I made an Aura with a Kill Effect , and played with the search Filters
was pretty easy actually not sure why i thought i had to do it with triggers.
the spawn type behaviour sounds cleaner doh ,
